Proposal Summary Shiga toxigenic E. coli are significant human pathogens with potentially fatal sequelae. Current treatments are limited to supportive care and antibiotics are contraindicated. The genetic determinants associated with severe human disease is poorly understood although it is correlated with the prophage content of the isolate. We have found that these prophage elements are unusually rich in sRNA regulators and may increase Shiga-toxin production. This proposal will provide a comprehensive, transcriptome-wide analysis of sRNA functions and phenotypes under infection-relevant conditions.
